Lytic Polysaccharide Monooxygenases as Chitin-Specific Virulence Factors in Crayfish Plague

چکیده

The oomycete pathogen Aphanomyces astaci, also known as “crayfish plague”, is an obligate fungal-like parasite of freshwater crustaceans and considered responsible for the ongoing decline native European crayfish populations. A. astaci thought to secrete a wide array effectors enzymes that facilitate infection, however their molecular mechanisms have been poorly characterized. Here, we report identification AA15 lytic polysaccharide monooxygenases (LPMOs) new group secreted virulence factors in astaci. We show this enzyme family has greatly expanded compared all other oomycetes, it may infection through oxidative degradation crystalline chitin, most abundant found crustacean exoskeleton. These findings reveal roles LPMOs animal–pathogen interactions, could help inform future strategies protection farmed endangered species.

Evolution of substrate specificity in bacterial AA10 lytic polysaccharide monooxygenases

BACKGROUND Understanding the diversity of lignocellulose-degrading enzymes in nature will provide insights for the improvement of cellulolytic enzyme cocktails used in the biofuels industry. Two families of enzymes, fungal AA9 and bacterial AA10, have recently been characterized as crystalline cellulose or chitin-cleaving lytic polysaccharide monooxygenases (LPMOs). Here we analyze the sequence...

Lytic polysaccharide monooxygenases disrupt the cellulose fibers structure

Lytic polysaccharide monooxygenases (LPMOs) are a class of powerful oxidative enzymes that breakdown recalcitrant polysaccharides such as cellulose. Here we investigate the action of LPMOs on cellulose fibers. After enzymatic treatment and dispersion, LPMO-treated fibers show intense fibrillation. Cellulose structure modifications visualized at different scales indicate that LPMO creates nickin...

The yeast Geotrichum candidum encodes functional lytic polysaccharide monooxygenases

BACKGROUND Lytic polysaccharide monooxygenases (LPMOs) are a class of powerful oxidative enzymes that have revolutionized our understanding of lignocellulose degradation. Fungal LPMOs of the AA9 family target cellulose and hemicelluloses. AA9 LPMO-coding genes have been identified across a wide range of fungal saprotrophs (Ascomycotina, Basidiomycotina, etc.), but so far they have not been foun...

Lytic polysaccharide monooxygenases from Myceliophthora thermophila C1 differ in substrate preference and reducing agent specificity

BACKGROUND Lytic polysaccharide monooxgygenases (LPMOs) are known to boost the hydrolytic breakdown of lignocellulosic biomass, especially cellulose, due to their oxidative mechanism. For their activity, LPMOs require an electron donor for reducing the divalent copper cofactor. LPMO activities are mainly investigated with ascorbic acid as a reducing agent, but little is known about the effect o...

Production of four Neurospora crassa lytic polysaccharide monooxygenases in Pichia pastoris monitored by a fluorimetric assay

UNLABELLED BACKGROUND Recent studies demonstrate that enzymes from the glycosyl hydrolase family 61 (GH61) show lytic polysaccharide monooxygenase (PMO) activity. Together with cellobiose dehydrogenase (CDH) an enzymatic system capable of oxidative cellulose cleavage is formed, which increases the efficiency of cellulases and put PMOs at focus of biofuel research. Large amounts of purified P...
